Montefiore and Einstein evaluate new drug combination to fight COVID-19
Montefiore Health System Albert Einstein College of Medicine has begun the next phase of the Adaptive COVID-19 Treatment Trial (ACTT) to evaluate treatment options for people hospitalized for severe COVID-19 infections. A new iteration of this trial, known as ACTT 2, is sponsored by the National Institute of Allergy and Infectious Diseases (NIAID), which is part of the National Institutes of Health.
In March, Montefiore first participated in a multicenter trial in New York to evaluate the intravenously administered broad-spectrum antiviral drug lemdesibir. The trial’s provisional results were announced last month, and on Friday New England Medical Journal, Patients with COVID-19 treated with lemdecivir showed a mean recovery of 11 days compared to 15 days for patients in the placebo group-statistically significant improvement. Of the 1,063 clinical trial participants, 91 (about 10%) came from Montefiore and Einstein.
Following the promising results of lemdecivir, this trial is currently investigating lemdecibir in combination with baricitinib or placebo in a double-blind, randomized trial. Baricitinib is marketed to reduce inflammation associated with rheumatoid arthritis. Researchers say that valicitinib in combination with lemdecivir can deadly overwhelm the lungs and other parts of the body of people with COVID-19 when the immune system responds to coronavirus infections. Dr. Zingman oversees the original lemdesibir study at Montefiore and directs ACTT 2.
Patients enrolled in the ACTT 2 have laboratory confirmed coronavirus infections, rattling sounds during breathing, need for oxygen supplementation, abnormal chest x-rays showing pneumonia, or need for ventilator, etc. Hospitalized for pulmonary complications. All patients will receive lemdecivir intravenously for up to 10 days. Half of the patients will also receive baricitinib orally and the other half will receive the same placebo for the same 14 days.
