Precise delivery of cargo within a cell?
A research team led by Professor Seo Dae-ha (President Lee Kun-woo) of the Department of Physical Chemistry at the Korea Advanced Institute of Science and Technology developed a new real-time microscope technology and succeeded in observing the behavior of “motor proteins.”[1]This may be key to elucidating efficient material transport strategies of cells. Using nanoparticle probes, high-resolution microscopes and Fourier transform algorithm techniques, the research team developed a “Fourier transform-based plasmonic dark-field microscope” (FT-pdf microscope) with positional and angular accuracy comparable to that of electron microscopes, achieving the highest level of existing optical microscopes.
Cells efficiently transport materials through intracellular vesicles called endosomes.[2]Materials are transported to their destinations by motor proteins that move along a complex microtubule network. Observing the movement and rotation of endosomes during transport provides important information for understanding how intracellular transport is efficiently regulated, and can help elucidate cell functions and diseases.
To visualize this transport process, the team developed an FT-pdf microscope that performs analysis by Fourier transform.[3] Technology using nanoparticles with “polar angle dependency”[4]By continuously imaging the scattering signal observed with circularly polarized light over a long period of time and combining it with existing single particle tracking technology, it is possible to observe the movement and rotation of particles in real time.
Using plasmonic dark-field microscopy, the team discovered temporal patterns in the rotational movement of endosomes within cells, which they interpreted as similar to the reinforcement learning strategies of navigation robots and internet search engines. Analyzing the real-time strategies of endosomal trafficking and applying them to disease cell models can help explain and diagnose the causes of disease.
“As shown in this study, normal cells that make up the human body appear to be equipped with the robotic data learning technology that humanity is actively developing,” said Professor Seo of DGIST's Department of Chemistry and Physics. “This molecular-level strategy holds the key to precise material transport and is another research topic. Our research findings are expected to contribute to the understanding and diagnosis of diseases in the future through their application to diseased cells,” he added.
The research was supported by the Ministry of Science and ICT, the National Research Foundation of Korea's Basic Research Program and Engineering Research Center (ERC) Program, DGIST's Grand Challenge Research and Innovation Project (D-GRIP), and the HRHR+ Program.
