Non-genetic theories of cancer address contradictions in the current paradigm

It's time for researchers to rethink the current cancer paradigm as a genetic disease, Sui Huang and colleagues of the US Institute of Systems Biology argued in a new essay published in the Open Access Journal on March 18th PLOS Biology.

A general theory of the origin of cancer is that otherwise normal cells accumulate genetic mutations that allow them to be unidentified. This paradigm has driven large-scale cancer genome sequencing projects, such as the Cancer Genome Atlas, identify cancer-driven mutations, and develop drugs designed to target aberrant proteins and pathways.

In their new essay, Huang and his colleagues argue that this theory of somatic mutation for cancer is counterproductive. They point to inconsistencies in sequencing data that are inconsistent with current theory, including the fact that many cancers do not have known driver mutations, but that some normal tissues can harbor cancer-causing mutations.

They propose a broader, more “overall” view that embraces biological biology and theory. Specifically, we encourage consideration of alternative paradigms involving non-genetic processes involved in tumorigenesis. For example, they explain the concept of cancer as a result of disruption of gene regulatory networks (Huang). The authors argue that these alternative explanations guide experiments and promote understanding of the origins of cancer.

The authors state that “a complete embrace of the idea that cancer origins go beyond the realm of genetic mutations will open up a new perspective on cancer treatment and prevention. If we accept that not all cancers are mutants, we can embrace strengthening public health policies aimed at preventing exposure to environmental non-customer factors that may promote nutritional supplements, such as working from home, such as promoting food products and other intakes such as work from home.
