Small molecules shaped like lasso could be a powerful tool in combating infectious diseases, according to a new study. Nature Co-authored by researchers at the University of Illinois Chicago.

Lariosidin is a newly discovered member of the Lassopeptide family. It is a small protein shaped like a lasso, with an amino acid loop at one end that passes through the tail. The new peptide was discovered in bacteria collected in the backyard of one of Canadian scientists.
After McMaster researchers observed that lariosidin can kill microorganisms that cause several diseases, they worked with researchers at UIC to study how it works. In biochemical and structural experiments, the team discovered that lariosidin binds and blocks ribosomes, the factory of cells that make new proteins.
“I found new jobs with these lassopeptides,” Travin said. “No one knew that lassopeptides could kill bacteria by binding to ribosomes and the bacteria were unable to make new proteins.”
Lariosidin binds to a different site from where other antibiotics bind to the ribosome, thus avoiding the antibacterial evolved defenses to resist other drugs.
“In the field of antibiotic discovery, we need weapons to kill by targeting something different from what we had before,” says Polikanov, an associate professor of biological sciences. “Otherwise, previously used protections automatically lead to protection against new molecules.”
The unique structure of the peptide may also help to avoid another common bacterial defense, Travin said. To tie the ribosome, antibiotics must first enter the bacterial cells. Many drugs sneak in through transporters, but bacteria can exchange or remove these to block the drug.
In contrast, lariosidin has a strong positive charge, allowing it to pass directly through the membrane without the need for a transporter. This makes the molecule a broad spectrum antibiotic.
“If you don't rely on a particular transporter, you can penetrate most of the bacteria,” Travin said. “And if you don't need a transporter, the probability of resistance is lower.”
Researchers have also studied a variant of lariocidin that takes on a more complex three-dimensional shape, looping its tail to resemble a pretzel. This even more stable structure could be the most promising candidate for clinical development, the researchers said. Bioinformatic analysis of available bacterial genomes suggests that there may be other lasso and pretzel peptides targeting ribosomes that have not yet been discovered in nature.
“Essentially, lariosidin is a founding member of a new family of antibiotics with similar mechanisms of action,” Travin said. “Time shows whether other peptides of this kind will become even more active than this. But we're already stepping into the door.”
