A new class of antibiotics found in soil samples

The process led to groups that exhibit broad spectral antibiotic activity, effective in killing both gram-positive and gram-negative pathogenic bacteria, and discovering the group along with mycobacteria that cause tuberculosis.

Lariosidine binds to the aminoacyl site of the ribosome, Wright says. “It is an acceptor site and contains transfer RNA containing new amino acids [tRNA] bind. And when lariosidin is bound to this site, the ribosome cannot properly read the codons of messenger RNA, so protein mistranslation is not an antibacterial compound.

Wright and the team followed the structural analysis of lariosidin in tests on human cell lines. Wright says this lack of toxicity to human cells is because human ribosomes are quite different from bacterial ribosomes that lariosidin cannot bind to them. The team then infected mice. “Despite 30 years of research into the discovery of lassopeptides, this study presents the first report on their in vivo testing,” says Crake. Finding success in mouse models, he adds, “to demonstrate the commercial potential of this class of peptides.”

Wright and his colleagues are currently investigating how modifying lariosidin produces other interesting antibacterial compounds. “I think we actually hit a gold vein looking for a new, ribosome-oriented antibiotic that's really novel, and we're really excited about it,” he says. However, it may still take some time for any of these potential drugs to enter a clinical setting.

Wright says the job would not have been possible without “cross-border collaboration between the University of Illinois University of Chicago group and team.” But the conflict between the US and Canada has created “unnecessary confusion” that complicates future collaborations, he says.

For now, Wright says that the discovery of Larioshidin and the mode of action through international collaboration are “a great example of how synergistic skills and research funding are gathered to solve some of these big problems we face as a society.”
