Single-dose Valxavir treatment can reduce influenza transmission

Arnold S. Mont, Maryland, and colleagues in Ann Arbor, University of Michigan's Department of Public Health, University of Michigan, examined the efficacy of single doses. Influenza-positive index patients ages 5-64 were randomly assigned to receive valvexavir or placebo within 48 hours of onset of symptoms (726 patients and 731 patients, respectively).
The researchers found that laboratory-confirmed influenza transmission was significantly lower in valoxavir than placebo by day 5 (adjusted incidence rate, 9.5 vs. 13.4%, adjusted odds ratio, 0.68; 95.38% confidence interval, 0.50 to 0.93; p = .01), adjusted relative risk reduction is 29%. By day 5, the adjusted incidence of influenza virus transmission that caused symptoms was 5.8 and 7.6% for valoxavir and placebo, respectively, but the difference was not significant (adjusted odds ratio, 0.75; 95.38% CI, 0.50 to 1.12; p = .16). During the follow-up period, the emergence of drug-resistant virus occurred in 7.2% of index patients in the valoxavir group. No resistant viruses were detected during household contact. There were no new safety signals reported.
