Experts find a link between risk factors for Alzheimer’s disease and severity of Covid-19
Researchers say that genes that increase the risk of Alzheimer’s disease may increase a person’s susceptibility and severity to Covid-19. This study, published in the journal CellStemCell, suggests that the gene ApoE4 was more susceptible to SARS-CoV-2 infection.
“Our study shows a causal link between the risk factors ApoE4 and Covid-19 for Alzheimer’s disease and explains why some, but not all, Covid-19 patients have neurological symptoms,” the United States said. Hope City researcher Yanhong Shi said. ..
“Understanding how risk factors for neurodegenerative disease affect the susceptibility and severity of Covid-19 successfully addresses Covid-19 and its potential long-term effects in different patient populations. “It helps,” Shi added. For this study, the team first used pluripotent stem cells (iPSCs) to create brain cells in the lab. This is a type of stem cell that can become virtually any type of cell.
Next, newly created neurons and astrocytes (a type of helper cell) were infected with SARS-CoV-2. They found that both cell types were susceptible to infection. The team then used iPSC to create a brain organoid, a 3D tissue model that mimics certain functions of the human brain.
They created one organ model with astrocytes and one organ model without astrocytes. Later, they infected both brain organoid types with the virus and found that those with astrocytes promote SARS-CoV-2 infection.
The team further studied the effect of ApoE4 on susceptibility to SARS-CoV-2 by generating neurons from iPSCs that were “reprogrammed” from cells in Alzheimer’s disease patients, including ApoE4. The team used gene editing to modify some of the ApoE4 cells created in iPS cells to contain the genotype ApoE3, which is considered neutral. Next, ApoE3 and ApoE4 iPSCs were used to generate neurons and astrocytes.
Both ApoE4 neurons and astrocytes were more sensitive to SARS-CoV-2 infection than neutral ApoE3 neurons and astrocytes.
In the final part of the study, researchers tested to see if the antiviral drug remdesivir inhibits viral infections of neurons and astrocytes. They found that the drug could normally lower viral levels in astrocytes and prevent cell death.
