Here’s how to stop a cold-induced toothache and hypersensitivity

In a recent study, scientists have found that odontoblasts, the cells that make up the dentin of teeth, function to sense the cold, which can cause toothache. They also found a way to block the path to cold-sensitive teeth.
Researchers report in Science Advances that they have discovered a new function of the shell under the tooth enamel that encloses the soft pulp, including odontoblasts, dentin-forming cells, and nerves and blood vessels. Pathologists Jochen Lennerz, MD, and PhD, chief author and one of the medical directors of the Massachusetts General Hospital’s Center for Integrated Diagnosis, said: (MGH). “This study brings a new function to this cell, which is exciting from a basic science point of view, but we also know how to interfere with this cold sensing function and control toothache.”
Teeth that are damaged by being exposed to a cold can occur for many reasons. Many experienced severe cold pain, for example, when a tooth was punctured from an untreated tooth decay. However, teeth can also be very sensitive to the cold caused by aging gum erosion. Some cancer patients treated with platinum-based chemotherapy have an extreme sensitivity to cold throughout the body. “Facial breeze is recorded as extreme pain in the teeth, and some patients may even discontinue treatment,” says Lennerz.
Tooth pain is notorious for being difficult to study. Tooth stiffness is a difficult tissue to study and requires the teeth to be opened to induce pain in human teeth. Therefore, the research team conducted experiments on mice in which molars were drilled under anesthesia. Mice with tooth injuries show pain in their behavior. For example, drink up to 300% more sugar water than your litter without injuring your teeth. In a previous study, a team of researchers discovered TRCP5, a protein encoded by the TRCP5 gene, which is expressed in nerves in many parts of the body. Their previous findings have allowed researchers to focus on TRCP5 as a mediator of cold pain.
By studying genetically modified mice that do not carry the TRCP5 gene, researchers found that mice with damaged teeth did not show increased drinking behavior and behaved like mice without tooth damage.
“There is conclusive evidence that the temperature sensor TRCP5 transmits cold air through odontoblasts, firing nerves and causing pain and cold hypersensitivity,” said Lennerz. “This sensitivity to cold may be a way for the body to protect damaged teeth from additional injuries.”
Specifically, in response to cold, the TRCP5 protein opens channels in the odontoblast membrane, allowing other molecules, such as calcium, to enter and interact with the cell. For example, if the pulp of a tooth becomes inflamed from deep caries, TRCP5 becomes excessive, causing an increase in electrical signals through nerves that exit the root of the tooth and run into the brain, where pain is perceived. As the gums recede from aging, the teeth can become hypersensitive because the odontoblasts sense the cold in the newly exposed areas of the tooth. “Most cells and tissues slow down metabolism in the presence of cold, so donor organs are placed on ice,” says Lennerz. “But TRPC5 makes cells more active in the cold, and the ability of odontoblasts to sense cold through TRPC5 makes this discovery very exciting.”
Lennerz confirmed the presence of the TRPCS protein in the extracted human teeth. This was a technical tour. “Our teeth are not intended to be cut into ultra-thin layers for microscopic examination,” said the TRPC5 channel, which first decalcifies the tooth, puts it in epoxy, and then slices it. Lennerz said he needed to identify. Odontoblasts.
The research team also identified pharmacological targets to minimize dental hypersensitivity to cold. For centuries, clove oil has been used as a remedy for tooth pain. The activator of oil of clove is eugenol, which happens to block TRCP5. Although toothpastes containing eugenol are already on the market, the results of this study may lead to more powerful applications for treating cold-sensitive teeth. There may also be new uses for eugenol, such as treating patients systemically for extreme sensitivity to chemotherapy. “We look forward to seeing how other researchers apply our findings,” says Lennerz.
