The researchers analyzed patient tumor biopsies taken before MAPKi treatment and at clinical relapse and found that the melanoma genome undergoes extensive “shatter and resuture” in response to treatment. Reconnects chromosomes so that they are circular extrachromosomal DNA (ecDNA). They found that cancer cells can produce and retain up to 100 copies of ecDNA.
