US healthcare workers who received a booster vaccine during the Omicron period (December 2021 to April 2022) had a 45% reduced risk of developing symptoms six weeks after COVID-19 infection, a research team announced. Study today in Open Forum Infectious Diseases.

The study findings came from a self-reported symptom questionnaire completed six weeks after symptom onset and were a subanalysis of data collected during the Preventing Emerging Infectious Diseases through Vaccine Efficacy Trials Project (Project PREVENT).

Researchers enrolled a total of 2,478 participants, of whom 1,422 (57%) had COVID-19. All participants were fully vaccinated with two doses of the COVID-19 vaccine, and 66.3% had received a third booster dose. All participants reported COVID-19-like symptoms within 14 days of the eligibility test.

The prevalence of symptoms at 6 weeks was 26% (373) in COVID-19-infected individuals and 18% (195) in non-COVID-19 individuals. In SARS-CoV-2-positive participants, fever or cough was the initial symptom in 71.9% of cases.

A recent booster vaccination reduces the chance of developing symptoms

Among participants who were infected with COVID-19, those who received a booster COVID-19 vaccine were less likely to have symptoms 6 weeks after infection than those who did not (adjusted odds ratio [aOR]0.55; 95% confidence interval [CI]0.43 to 0.70).

If the third vaccination was administered within 16 weeks of infection, HCWs had lower odds of reporting respiratory symptoms (aOR, 0.69; 95% CI, 0.46-0.99), psychiatric symptoms (aOR, 0.68; 95% CI, 0.47-0.99), or any symptoms (aOR, 0.68; 95% CI, 0.53-0.87) compared with HCWs who received the vaccine more than 16 weeks earlier.

COVID-19 vaccination is effective in reducing the severity of acute illness and may therefore lead to a reduction in symptoms for six weeks in patients with symptomatic infection.
