Twice-yearly lenacapavir treatment prevents HIV infection in young women
HealthDay News—For adolescent girls and young women, a dose of lenacapavir given every 26 weeks is effective in preventing acquired HIV infection, according to a study published online July 24. Linda Gayle Becker, MB, ChB, PhD, from the Desmond Tutu HIV Centre at the University of Cape Town in South Africa, and colleagues conducted a randomly assigned phase 3 trial in adolescent girls and young women. Subcutaneous lenacapavir Participants received either daily oral emtricitabine-tenofovir alafenamide (F/TAF) or daily oral emtricitabine-tenofovir disoproxil fumarate (F/TDF, active comparator) in a 2:2:1 ratio every 26 weeks (2134, 2136, and 1068 participants, respectively). The relative efficacy of lenacapavir and F/TAF was assessed by comparing the incidence of HIV infection in the screened population (8094 participants) with estimated background incidence, and the relative efficacy was compared with F/TDF.
The researchers confirmed that 55 of 5,338 initially HIV-negative participants acquired HIV infection (0.0, 2.02, and 1.69 infections per 100 person-years in the lenacapavir, F/TAF, and F/TDF groups, respectively). In the screened population, the background HIV incidence rate was 2.41 per 100 person-years. HIV incidence with lenacapavir was significantly lower than background HIV incidence and HIV incidence with F/TDF (incidence rate ratios were both 0.00). No significant differences were found between HIV incidence with F/TAF and background HIV incidence, and no meaningful differences in HIV incidence were found between F/TAF and F/TDF.
“Twice-yearly lenacapavir offers a highly effective, unobtrusive option that has the potential to improve the use of pre-exposure prophylaxis in women,” the authors write.
Several authors disclosed ties to pharmaceutical companies, including Gilead Sciences, which makes lenacapavir and funded the research.
