Alzheimer’s patients don’t perceive pain in the same way — ScienceDaily
A new study from the Institute of Psychiatry, Psychology and Neuroscience (IoPPN) at King’s College London shows that pain signals are processed in the same way as healthy mice in a mouse model that mimics Alzheimer’s disease (AD). turned out not to be.
This research Nature Communications, suggested that pain perception may be altered in people with Alzheimer’s disease and questioned whether altering pain management in people with Alzheimer’s disease would improve their quality of life.
Chronic musculoskeletal pain is common in patients with Alzheimer’s disease but is largely untreated because it may go unreported due to the cognitive impairment associated with the disease.
In this study, researchers sought to investigate whether there were also changes in the body’s response to pain by the nervous system of people with Alzheimer’s disease.
In healthy mice, pain signals are transmitted from the point of origin to the central nervous system, initiating an immune response. The protein galectin-3 has been shown to be involved in pain signaling to the spinal cord. Once in the spinal cord, it binds to another protein, TLR4, and initiates an immune response.
In this study, researchers used an AD mouse model and administered blood to rheumatoid arthritis, a chronic inflammatory disease. They observed an increase in allodynia, pain caused by stimuli that normally do not cause pain, in response to inflammation. They also found microglia, resident immune cells in the spinal cord, and increased their activation. They determined that these effects were regulated by TLR4.
The researchers found that immune cells in the central nervous system of Alzheimer’s mice lack TLR4, so they cannot sense signals and respond to pain in the usual way.
As a result, mice with Alzheimer’s disease experienced less pain associated with joint inflammation, and immune cells responded less to pain signals received by the central nervous system.
Professor Marzia Marcanjo, Professor of Neuropharmacology at King’s IoPPN and lead author of the study, said, “Nociceptive pain, pain that is the result of tissue damage, is the second most common comorbidity in patients with Alzheimer’s disease. Our study showed that mice with Alzheimer’s disease lack TLR4, a protein essential for immune response processes in the central nervous system, thus impairing the body’s ability to process pain. change.
“These are important findings because untreated pain can contribute to the psychiatric symptoms of this disease. As research advances and our understanding of this area improves, more effective treatments will emerge.” , which could ultimately improve people’s quality of life.”
“The results of this study identify galectin-3/TLR4 as a potential therapeutic target for chronic pain,” said George Sideris-Lampretsas, a Ph.D. student at King’s IoPPN and lead author of the study. “It can not only do, but it can have an impact,” he said. Most important, however, is raising awareness of the underreported and untreated pain experienced by AD patients. “
